PT-141
A melanocortin agonist studied for sexual desire and function.
Overview
PT-141 (Bremelanotide) is a synthetic cyclic peptide that activates melanocortin receptors in the brain. It is FDA-approved (as Vyleesi) for hypoactive sexual desire disorder (HSDD) in premenopausal women and is widely researched for its effects on sexual arousal and libido in both sexes.
Dosing
Injection
- Starting dose
- 0.75 mg
- Common dose
- 0.75–1.75 mg
- Frequency
- As needed (max 1–2 doses/day)
- Notes
- Approved dose is 1.75 mg subQ; start low to limit nausea.
Nasal

Reported research/reference doses for educational purposes only. Always verify against primary literature and applicable regulations. Not medical advice.
How it works
PT-141 acts centrally on melanocortin-3 and melanocortin-4 receptors (MC3R/MC4R) in the paraventricular nucleus of the hypothalamus. This signaling pathway modulates sexual arousal and desire independent of the cardiovascular system, so it works even in the presence of erectile dysfunction.
Common research uses
- Female hypoactive sexual desire disorder
- Sexual arousal and libido research
- Erectile function studies
- Central melanocortin pathway research
Considerations
Prescription medication. Common side effects include nausea, flushing, and transient blood-pressure changes. Not for use with nitrates or uncontrolled hypertension.
